The enzymes utilize cytosolic UDP-glucuronic acid UDP-GlcUA ; and UDP-NAcetylglucosamine UDP-GlcNAc ; as donors to produce the linear hyaluronan polymer containing up to 25, 000 repeating disaccharides with the structure [GlcUA1- 3GlcNAc1- 4]n 24 ; . Hyaluronan synthesis is known to be controlled at the level of Has transcription 25 ; , which is influenced by growth factors, cytokines and hormones 2, 3, 25-30 ; . There is also evidence for posttranscriptional factors like HAS protein phosphorylation that may contribute to the rate of hyaluronan synthesis 31, 32 ; . The expression of UDP-glucose UDP-Glc ; dehydrogenase UGDH ; which converts UDP-Glc to UDP-GlcUA, is often upregulated at the same time as hyaluronan production increases 33 ; , and experimental increase and decrease of UGDH expression exerts corresponding changes in hyaluronan synthesis 34 ; . This suggests that the rate of hyaluronan synthesis may also be regulated by the level of HAS substrate concentration. Indeed, 4methylumbelliferone 4-MU ; as a good substrate of glucuronidation consumes UDP-GlcUA and inhibits hyaluronan synthesis 35 ; . Many delivery systems are available for medications approved for asthma treatment: oral tablets, nebulizers, MDIs, and dry powder inhalers DPIs ; . SUMMARY Keratinocyte growth factor KGF ; activates keratinocyte migration and stimulates wound healing. Hyaluronan, an extracellular matrix glycosaminoglycan that accumulates in wounded epidermis, is known to promote cell migration, suggesting that increased synthesis of hyaluronan might be associated with the KGF response in keratinocytes. Treatment of monolayer cultures of rat epidermal keratinocytes led to an elongated and lifted cell shape, increased filopodial protrusions, enhanced cell migration, accumulation of intermediate size hyaluronan in the culture medium and within keratinocytes, and a rapid increase of hyaluronan synthase 2 Has2 ; mRNA, suggesting a direct influence on this gene. In stratified, organotypic cultures of the same cell line, both Has 2 and Has3 with the hyaluronan receptor CD44 were upregulated and hyaluronan accumulated in the epidermis, the spinous cell layer in particular. At the same time the expression of the early differentiation marker keratin 10 was inhibited, while filaggrin expression and epidermal permeability were less affected. The data indicate that Has 2 and Has3 belong to the targets of KGF in keratinocytes, and support the idea that enhanced hyaluronan synthesis acts an effector for the migratory response of keratinocytes in wound healing, while it may delay keratinocyte terminal differentiation and hyaluronan.
